How nicotine in tobacco is measured, and what the number actually means

A nicotine percentage written into a tobacco contract looks like a property of the goods, in the way that net weight or bale count is a property of the goods. It is not. It is a laboratory result, produced by one lab, from one sample, on one day, and reported on one basis. Buyers who understand that read the figure differently, and they ask for different things before agreeing a price.

A nicotine figure is a result, not a fixed property

Nicotine is an alkaloid the tobacco plant makes and stores in the leaf. How much ends up there depends on the variety, the soil, the rainfall, the position of the leaf on the stalk, the curing and how long the material has sat since. Two fields of the same variety in one district can differ, and leaf from the top of the plant differs from leaf at the bottom by more than that.

A commercial lot is an aggregate of many plants, many plots and often many weeks of processing, so the single figure on the paperwork is a summary of a spread, not a constant. The useful question is not whether it is exactly right, but how well it represents the tonnage behind it.

Wet chemistry and instrumental methods

The trade uses two broad families of method, and they answer slightly different questions.

Wet chemistry works by separating the alkaloid from the leaf and quantifying it through a chemical reaction. These methods are long established, cheap to run and need no costly instrument. That last point matters commercially more than it sounds: a test cheap enough to run on every batch tells a buyer far more than an expensive test run on one lot in fifty.

Instrumental methods use chromatographic and spectroscopic equipment to separate compounds and measure them individually. They can distinguish nicotine from the minor alkaloids alongside it and work well at low concentrations, but they cost more per sample and per machine, so in commercial trade they tend to be used for referee results, regulatory work and research rather than routine lot control.

The practical consequence is worth holding on to. Wet chemistry generally reports total volatile alkaloid expressed as nicotine, which reads a little higher than nicotine alone. Instrumental work can report nicotine on its own. Neither is wrong. They are measuring slightly different things, and two figures produced by two families of method are not automatically comparable.

A figure without a stated basis is incomplete

Tobacco takes up and gives off moisture with the weather. The same material weighs more in a humid month than in a dry one, and since nicotine is reported as a percentage of weight, the percentage moves with the water even though the alkaloid has not changed.

This is why results are quoted on a dry basis or as received. A lot at a given percentage dry basis and a lot at the same percentage as received are not the same purchase, and the gap between them is water. When a buyer and a seller argue about an assay, the argument is very often about basis rather than about tobacco. A figure quoted without a basis is unfinished information.

Sampling drives accuracy more than the instrument does

A lab works with a few grams. Those grams stand in for many tonnes. If they do not represent the lot, no amount of instrument precision rescues the result, and the failure is invisible afterwards because the report looks perfectly normal.

Scrap, dust and rava make this harder than baled leaf. Fines and heavier mineral matter settle, lighter fibre rides up, and bags filled at different points in a week are not identical. A sample drawn from whatever stood nearest the door describes those bags and nothing else.

So the question to put to a seller is not only which method stood behind the figure, but what the sample represented. A seller comfortable with a jointly drawn sample is telling you something about the material. So is one who is not.

Ask for a figure tied to the lot, not to the leaf type

Published tables give nicotine ranges by tobacco type and growing region. They are useful for orientation and useless for pricing, because the ranges are wide and an average for a type says nothing about the specific tonnage on offer this season.

What a buyer should price against is a result from the actual lot, with the test date and the basis stated, and a clear account of what was sampled. That turns a number on a quotation into something that can be checked and compared with the last delivery.
